PCR ini banyak digunakan dalam berbagai penelitian dibidang molekuler untuk memperbanyak sekuen [[DNA]]. Enzim Pfu DNA polimerase mampu memperpanjang rantai DNA dalam suatu siklus. Sumber utama enzim ini adalah &amp;#039;&amp;#039;Pyrococcus furiosus&amp;#039;&amp;#039;. Keunggulan utamanya dibandingkan dengan enzim [[DNA polimerase]] lainnya ialah [[waktu paruh]]nya yang sangat lama dan adanya mekanisme perbaikan rantai DNA yang salah (&amp;#039;&amp;#039;proofreading&amp;#039;&amp;#039;). Selain itu, enzim ini bersifat termostabil atau bersifat tahan suhu tinggi.&lt;br /&gt;

Mekanisme &amp;#039;&amp;#039;proofreading&amp;#039;&amp;#039;-nya adalah 3&amp;#039;-5&amp;#039; eksonuklease. Melalui mekanisme &amp;#039;&amp;#039;proofreading&amp;#039;&amp;#039;, suatu kesalahan penambahan [[basa]] DNA saat amplifikasi dapat diperbaiki. Aktivitas &amp;#039;&amp;#039;proofreading&amp;#039;&amp;#039; ini memiliki keakuratan yang sangat tinggi sehingga enzim ini sering kali digunakan dalam reaksi pemanjangan [[primer]], [[kloning]], dan amplifikasi rantai DNA ujung tumpul (&amp;#039;&amp;#039;blunt end&amp;#039;&amp;#039;).&lt;br /&gt;
